Isolation refers to how "well-surrounded" a building is, usually referring to its walls and ceiling. A good isolation helps block out noise caused by doing some tasks or jobs. Adding spaces for doorways (which are necessary for citizens to enter the building) will not decrease isolation. If any walls are removed from a building, isolation will decrease.

Walls added to a building that aren't available by default to said building can still increase isolation. In other words, disabling the "auto-build walls" option for a building, therefore constructing only the building's interior, then constructing walls around the building will increase isolation in the same as if the "auto-build walls" option was enabled.
